KeyTec® TR-FRET BTK WT / CRBN PROTAC Binding Assay Kit can be used to screen PROTAC molecules that mediate the formation of a ternary complex between wild-type BTK protein and the CRBN/DDB1 complex. This kit is a binding immunoassay developed using KeyTec® TR-FRET technology, featuring simplicity, speed, high accuracy, and excellent reproducibility.
The basic principle of this method is as shown in the figure below. The KeyTec® TR-FRET Solar Eu1‑conjugated anti‑Tag1 antibody specifically recognizes the Tag1‑BTK WT protein, while the KeyTec® TR‑FRET LA2‑conjugated anti‑Tag2 antibody specifically recognizes the Tag2‑CRBN/DDB1 protein complex. The Tag1‑BTK WT protein and the Tag2‑CRBN/DDB1 protein complex can form a ternary complex mediated by the positive reference PROTAC molecule MT‑802, bringing Solar Eu and LA into close proximity. Upon excitation by an external light source, energy resonance transfer occurs between the donor and acceptor. Detection of signal intensity at a specific wavelength (665 nm) allows determination of the PROTAC molecule‑mediated binding between BTK WT and CRBN/DDB1.
